Activity 1: Conservation of plant genetic resources

ACTIVITY DESCRIPTION

(1) Defining conditions for maintaining varieties in isolated conditions of an „insect free“ greenhouse in which the virus free material will be maintained in a sustainable way and with no risk for reinfection.

(2) Research on the efficiency of current virus elimination protocols based on meristem culture compared to cryotherapy; research related to the development of cryopreservation protocol for grapevine varieties.

Relation to the goals: Activity is connected to the goal 1 in the part which is related to the establishment of plant genetic resources collection to improve a model for safe grapevine varieties preservation in controlled conditions, by eliminating vegetative transmitted plant viruses and by the development of cryopreservation protocol for grapevine varieties.

Activity 2: Phenotyping

ACTIVITY DESCRIPTION

(1) determination of composition and content of polyphenols, volatile compounds and proteins responsible for their synthesis in grapevine varieties.

(2) Research the difference between indigenous grapevine varieties on downy mildew resistance.

Relation to the goal: Activity is connected to the goal 1 in the part which is related to the collection phenotyping in order to define the values of grapevine plant genetic resources conservation for production, and for further breeding as well.

Activity 3: Genotyping 

ACTIVITY DESCRIPTION

(1) Research related to genome stability evaluation during cryopreservation and cryotherapy of grapevine varieties.

(2) Establishing a platform for varieties identification and determination of intervarietal variability of the most important indigenous varieties.

Relation to the goals: Activity is connected to the goal 1 in the part which is related to the collection genotyping in order to define the influence of cryopreservation on genetic stability of preserved material. Accurate identification by molecular markers is the basic condition for efficient preserving indigenous grapevine varieties. Intravarietal variability in indigenous cultivars evaluation is essential for their preservation.
